An improved isolation method for the cultivation of the crayfish plague fungus, Aphanomyces astaci

Abstract
Experienced personnel using a well-equipped laboratory can even today find difficulties in isolating the crayfish plague fungus, Aphanomyces astaci. Contamination with bacteria is the main problem in isolation and cultivation of A. astaci in vitro. Two methods of isolation are compared in this paper. Method 1 follows the technique described by Cerenius et al. (1988), whereas Method 2, a modified method, follows a combination of techniques used by Cerenius et al. (1988) and Alderman and Polglase (1986) and involves new steps. Method 2 was more successful than Method 1: 24% of A. astaci-like moulds were cultivated using Method 1 compared with 70% using Method 2. The prevailing problem with Method 1 was bacterial contamination, whereas in Method 2 bacterial contamination was seen only in one case out of 23. The modified method offers a technique that increases the chance of successful isolation of the crayfish plague fungus.
